OCT4 is a key transcription factor that plays an important role in embryonic development. The human OCT4 gene can produce at least four protein isoforms, namely OCT4A, OCT4B-265, OCT4B-190 and OCT4B-164. Among them, OCT4A plays a role in maintaining the characteristics of stem cells and is the most studied isomer. Previous studies have proved that OCT4B-190 can play a role in the process of cellular stress, and the function of other isomers of OCT4B is not yet clear.
Research by Dr. Dai Jianwu from the Institute of Genetics and Developmental Biology, Chinese Academy of Sciences found that OCT4B-265 is up-regulated under genotoxic stress (such as mitomycin C, doxorubicin treatment or ultraviolet radiation). Molecular and biochemical studies have shown that the response of OCT4B-265 to genotoxic damage only appears in some cells with stem cell characteristics. Under genotoxicity, OCT4B-265 can promote apoptosis, and this effect is dependent on p53.
The research results reveal the new function of OCT4B protein, which has important reference value for understanding the diversity of OCT4 gene protein types and the complexity of functions. At the same time, OCT4B-265 protein may have important application prospects in preventing stem cell genotoxic damage.
